The Effects of Tolbutamide on the Renal Fine Structures in Alloxan Induced Diabetic Rabbits


Abstract
The author sudied the effects of tolbutamide on thn fin stru~.ures of th a}loxan-diab~tic rabbt hidne}-. Diabetes was induced by The administration of alloxan 130 mg per kg. of body eight to the rabbits. And then the diabetic animals were administered 100mg and. 200 mg per kg. of body ~~e}ghts of tolbutamide for 3 and G months, respectively.
The results were
1. Light microscopical}y, congetion and partial formation of thrombus in tike glomeruli, and vacnolar degeneration in the renal tubules were observed in both alloxan-diabetic and tolbutamide ec::,inis~er:d rabbits. i3ut th<~ changes were mild.
2. Elciron microscopicrcally, alloxan-diabetic rabbit renal gioneruli showed partial fusion of the for precesses, imgula:ities of the endothelial margins, hyp~rplasia of tli~~ metianrial ma*rig. ~~-ith inci~~ase of basment membrane Iike fibrils within the matrix. and increasc of collagen fibers an~i bwszmen¢¥: membrane like fibrils within the ;n:sangial ells. The proximal convluted tubular cells snowed swelling and irregular arrangement of the mitochonc?ria, increase of the lysosomes, anc! irregularity of the micrnvilli. All of the changes were mild, but they wre mere marked in the 6 month administered group than in the 3 month group. :[n the tolbutamide administered diabtic animals, more marked similiar charges were also observed. In addition to the above changes, re tention of electron dense materials within the mesangial matrix, and marked dilatation of the ER of :.lesangial cells were also observed in tolbutamide treated animals. Thos changes were more severe in high dosage administered animals.
3. With PAM staining, collagen fibers in the mesangial cells and PAM positive materials in the proximal convoluted tubular cells were confirmed.
4. According to these findings, toltutamide treatment o;z the alloxan-diabtic rabbits may inducthe changes of mesangium, because alloxan-diabetes alone d.o not make such changes. Since these cells are intimately related to hyprtension, thes findings are to by very important.
